Quercetin with Antioxidant Activity Inhibits Matrix Metalloproteinase-2 and -9 in HT1080 Cell Line

Abstract

Quercetin is a plant-derived bioflavonoid that has been used as a medicinal supplement for anti-allergy, anti-cancer and antioxidant. Here, we utilized in vitro monitoring lipid peroxidation and gelatin zymography assay to analyze its antioxidant activity related to tumor invasion in human fibrosarcoma cells (HT1080) exposed to hydroxyl radical. Quercetin exhibits the antioxidant effect on 1,1-diphenyl-2-picrylhydrazy (DPPH) free radical at above 4¥ìM as well as the inhibitory effect on lipid peroxidation in vitro, but not in live cells. In addition, quercetin inhibits not only both MMP-2 and MMP-9 expressions but also them under phorbol 12-myristate13-acetate (PMA) stimulation, with the increase of tissue inhibitor of MMP (TIMP-1) expression. Thus, Quercetin mediates the interaction between anti-oxidation activity and expression of MMPs in cooperation to inhibit tumor invasion. These findings suggest that quercetin can be applicable to chemoprevention of tumorigenesis.
